紫外-可见分光光度法测定汉麻果胶中总皂苷的含量

Content determination of total saponin in China-Hemp Pectin by ultraviolet spectrophotometry

目的建立汉麻果胶中总皂苷含量的测定方法。方法以蕨麻苷Ⅱ号为对照品,5%香草醛-冰醋酸和高氯酸的混合溶液为显色剂,采用紫外分光光度法在500 nm处测定汉麻果胶总皂苷的含量。结果蕨麻苷Ⅱ号检测浓度在70～177μg与吸光度呈良好的线性关系(r=0.999 3);平均加样回收率为99.0%,RSD=1.7%(n=6)。结论本方法简便,可操作性强,灵敏度高,是汉麻果胶总皂苷含量测定的一个切实可行的方法,为汉麻果胶有效部位的进一步研究提供依据。

Objective To establish a content determination method for total saponin in China-Hemp Pectin. Methods The content was determined with 5 % vanillin-glacial acetic acid perchloric acid as color developing reagent and JMG II was used as the control sample. The detection wavelength was 500 nm by ultraviolet spectrophotometry. Results The linear range of JMG II was 70-177 ug （r=0. 9993）, and the average recovery was 99.0%, RSD=1. 7% （n=6）. Conclusion This method is valid, practicable and reliable for the research and development of China-Hemp Pectin.
