摘要
目的建立高效液相色谱测定小儿至宝丸中胆红素含量的方法。方法采用HPLC法进行测定,色谱柱为Dikma C18(250 mm×4.6 mm,5μm);流动相:为氯仿-甲醇-2%二乙胺溶液(80∶19∶1),检验波长:450 nm,流速:1 mL.min-1,柱温:45℃。结果胆红素能完全分离,胆红素在浓度0.782 5~4.695μg.mL-1与峰面积呈良好的线性关系,回归方程Y=2.659×104X-156.9,r=0.999 9,加样回收率为99.2%,RSD为1.0%(n=9)。结论本方法可用于测定小儿至宝丸中胆红素的含量,方法简便,准确可靠。
Objective To establish an HPLC method for the determination of hilirubin in Xiaoer Zhibao Wan. Methods HPLC was used. The separation was performed on a Dikma C18 (250 min×4.6 mm, 5 um) column. The mobile phase consisted of chloroform- methanol-2% diethylamine solution (80 : 19 : 1) . The detection wavelength was 450 nm. The flow rate was 1.0 mL · min-1 and the column temperature was 45 ℃. Results The calibration curves of bilirubin showed good linearity over 0. 782 5-4. 695 ug ·mL- 1, and the regression equation was Y= 2. 659 × 10^4 X- 156.9, r=0. 999 9. The average recovery was 99. 2%, and RSD was 1.0%. Conclusion The method is simple, accurate and reliable, which can be used to determine bilirubin in Xiaoer Zhibao Wan.
