白藜芦醇对束缚应激大鼠血小板L-精氨酸/一氧化氮通路的影响 被引量:3

摘要 目的探讨白藜芦醇对急性应激大鼠血小板聚集功能和L-精氨酸/一氧化氮合酶/一氧化氮通路的保护作用及机制。方法复制水浸-束缚应激(WRS)大鼠模型,分对照组,应激组,白藜芦醇低、中、高剂量组,白藜芦醇各组于应激前分别按30、601、20mg/kg灌胃;对照组和应激组灌服同等容量的生理盐水。应激8h后测定血小板最大聚集率、血小板孵育液中亚硝酸盐(NO2-)含量、血小板NOS活性和L-Arg转运。结果白藜芦醇明显抑制WRS时血小板最大聚集率的升高,缓解WRS所致的血小板L-Arg转运、NOS活性和孵育液NO2-含量下调,以白藜芦醇中剂量组作用显著(P<0.05或P<0.01);person相关分析,白藜芦醇对WRS大鼠血小板最大聚集率的影响与NO释放呈显著负相关,r=-0.808 7,P<0.01。结论白藜芦醇可抑制WRS刺激的血小板聚集,改善应激损害的L-Arg/NO通路功能,两者的改变呈负相关。 Objective To investigate the effects of resveratrol on rats' platelet function and L-Arg/NOS/NO pathway under water-immerson restraint stress(WRS) condition and its mechanism.Methods The copy rat model under WRS condition was divided into control group,WRS group,low dose resveratrol group,middle dose resveratrol group and high dose resveratrol group.In control group and WRS group,rats were gavaged with the same dose hysiological saline.For others,rats were lavaged with 30,60 and 120 mg/kg resveratrol before WRS affect.After 8 hours WRS affect,a measurment for platelet maxium aggregation was taken by ADP induced and determined the content of NO-2incubated buffer of platelet、NOS activity and L-Arg transfer of rat platelets. Results The resveratrol could significantly restrain the increase of maxium platelet aggregation by ADP induced,relieve the LArg transfer、NOS activity and NO production of rats platelet during the WRS period.Middle dose resveratrol group showed the best result(P0.05 or P0.01).The platelet maxium aggregation showed the negative correlation with NO release by resveratrol in WRS rat′s platelets by person analysis,r=-0.808 7,P0.01.Conclusion The resveratrol can inhibit platelet aggregation during WRS,improve L-Arg/NO pathway functions by WRS damage,which has negative correlations between those changes.
