胰岛素样生长因子-1对实验性动脉粥样硬化斑块易损性的影响 被引量:2

摘要 目的探讨IGF-1对实验性动脉粥样斑块易损性的影响。方法 24只雄性新西兰兔,高脂饮食配合球囊拉伤腹主动脉建立动脉粥样硬化模型,将动物随机分为对照组、模型组、IGF-1干预组、辛伐他汀干预组。12周后处死动物,测血脂,取腹主动脉切片、染色,测量相关指标。结果辛伐他汀组与模型组间血脂TC、TG、HDL、LDL水平差异均有统计学意义,IGF-1组与模型组间血脂TG、HDL水平差异有统计学意义。模型组与正常对照组血脂水平差异有统计学意义。斑块顶部纤维帽厚度、肩部纤维帽厚度及斑块肩部、顶部帽/核比值,IGF-1组及辛伐他汀组与模型组比较,差异均有统计学意义,IGF-1组与辛伐他汀组比较,三项指标间差异无统计学意义。斑块脂核面积/斑块面积、斑块偏心指数以及斑块直径/对应管腔直径比较,IGF-1组、辛伐他汀组与模型组比,三项指标间差异均有统计学意义,IGF-1组与辛伐他汀组比较,三项指标间差异无统计学意义。结论采用高脂饲料喂养联合球囊扩张术建立的动脉粥样硬化模型,与人类粥样斑块的基本成分相似;IGF-1在一定程度上能够改善动脉斑块的易损性。 Objective To investigate the effects of IGF-1 in atherosclerosis rabbit model.Methods 24 New Zealand Rabbits were established atherosclerosis model by femoral artery angioplasty operations.All rabbits were divided into four groups:control group,model group,IGF-1 group and Simvastatin group.12 weeks later,executing all the rabbits,and measuring the blood lipid level.To take aorta and take path morphology observation and measure the related indicators.Results The differences about the TC、TG、HDL and LDL level among the simvastatin group and the model group were significance.There were significant differences in TG and HDL between the groups of IGF-1 and model.Blood-lipid level differences between the model group and the nomal group were statistical significance.To compare the group of IGF-1,simvastatin and model,the differences on top fiber cap thickness、shoulder fiber cap thickness and the ratio of fiber cap vs.Lipid core were statistical significance,however,the differences between IGF-1 and simvastatin group were non-statistics significance.To compare the group of IGF-1,simvastatin and model,the differences on lipid core area vs.plaque area,IE,and the degree of lumen relatively narrow,were statistical significance,however,the differences between IGF-1 and simvastatin group were non-statistics significance.Conclusions Rabbit atherosclerotic plaque model established with high-fat diet and femoral artery angioplasty operation contained main cell and matrix components of human lesions is similar to that of human being,and the factor of IGF-1 can improve the atherosclerotic plaque's vulnerability at a certain extent.
