摘要 大豆花叶病毒(Soybean mosaic virus,SMV)是马铃薯Y病毒组(Potyvirus)成员之一,大豆花叶病毒病可造成大豆10%~30%的产量损失,并严重影响大豆的品质。不同株系间致病力差异源于它们在碱基序列上的差异。采用传统的抗病育种技术育成的抗病品种抗谱窄,品种抗性可因病毒株系的变异而丢失。依靠RNA引发的基因沉默改善植物的抗病毒能力是基于RNA i(RNA inference)原理建立的植物抗病毒新策略,本研究对来源于武汉SMV分离物的CP基因和Nib基因进行了克隆,通过对保守区域的扩增,采用Gateway技术构建了大豆抗花叶病毒的RNA干扰载体。为防治大豆花叶病毒新技术的探索奠定基础。 Soybean Mosaic Virus(SMV) is belonging to Potyvirus.The disease caused by SMV is very popular,results in 10%-30% yield loss and exerts heave impact on soybean yield and quality.The pathogenicity of different virus strains varied with their nucleotide sequences.The utility of resistant varieties is the most economic pathway to control SMV.The resistant lines derived from conventional breeding material have narrow resistance spectrum,the resistance loss easily with the mutation of SMV strains.Acquired resistance based on RNA inference(RNAi) is a novel strategy to improve the plant resistance to virus.In this study,SMV isolate from Wuhan field was made sequence analysis,CP and Nib fragment were isolated based on homologous PCR,the fragment in most conserved region was chosed to construct RNAi vectors by use of gateway technique.The construct will be used in further transformation to improve soybean resistance to SMV.
