摘要 采用单因素实验和Box-Behnken响应面实验设计联用的方法,筛选和优化植物乳杆菌NCU116真空冷冻干燥的保护剂配方。单因素实验结果表明:海藻糖、D-山梨醇和脱脂奶粉3种保护剂效果好,能够显著提高菌种对冷冻干燥环境的耐受能力。响应面实验以上述3种保护剂为自变量,细胞存活率为响应值,结果表明:以脱脂奶粉、海藻糖和D-山梨醇的浓度分别为9.24%、4.47%和13.42%作为冻干保护剂配方进行验证试验,细胞存活率达到91.76%±1.82%,与理论预测值相符。 The protective agent formula of freeze-drying for Lactobacillus plantarum NCU116 was optimized by single factor experiment and a three-factor Box-Behnken design.The results of single factor experiment indicated that trehalose,D-sorbitol and the skim milk had significant effect to the survival of freeze-drying cells.The critical factors selected for this investigation were the skim milk,trehalose,and D-sorbitol,the cell survival rate used as response value.The optimal formula was skim milk concentration 9.24%,trehalose concentration 4.47% and D-sorbitol concentration 13.42%.The efficiency of protective agent formulation optimzed has been tested.The result showed that the cells survival rate reached 91.76%±1.82% in freeze-drying process,it more in line with the theoretical prediction.
