摘要
目的探讨激素性股骨头缺血坏死的发病机理及土鳖虫防治该病的作用机制。方法通过大剂量激素诱导体外培养的骨髓间充质干细胞成脂分化,同时给予土鳖虫含药血清干预。检测干预6天后细胞内成脂标志物PPARγmRNA和aP2mRNA的表达。结果土鳖虫含药血清可对抗激素诱导下的BMSCs PPARγmRNA和aP2mRNA表达的增加。结论土鳖虫防治激素性股骨头缺血坏死的机理不仅是改善股骨头的微循环,同时还与其抑制激素诱导下的BMSCs成脂分化有关。
Objective To explore the pathogenesis of hormonal avascular necrosis of femoral head and the therapeutic mechanism of Ground Beetle.Methods The differentiation of bone marrow mesenchy mal stem cells(BMSCs) into adipocytes in vitro were induced by large dose of steroid and treated simultaneously with Ground Beetle drug-containing serum for 6 days,then the expression of adipogenic markers,including PPARγ mRNA and aP2 mRNA,were detected.Results Ground Beetle could antagonize the increasing of expression of PPARγ mRNA and aP2 mRNA in steroid-induced BMSCs.Conclusion Ground Beetle can be used to treat hormonal avascular necrosis of femoral head and its mechanism is probably related with the effect of improving minicirculation of femoral head and inhibiting the adipogenic differentiation of BMSCs induced by steroid.
