甘草愈伤组织继代培养中激素浓度组合的优化研究 被引量:1

摘要 目的:寻找甘草愈伤组织继代培养中激素浓度的最佳组合。方法:以甘草种子为外植体,以无菌苗下胚轴及胚根诱导的初代愈伤组织为材料,利用二次正交旋转组合没计方法进行数学模拟,建立数学模型;并进行激素浓度组合寻优;在最佳培养条件下进行愈伤组织生长动力学研究。结果:模型的决定系数R2为0.9161;甘草愈伤组织继代培养中激素浓度的最佳组合为6-BA 1.514 mg/L+NAA 2.059 mg/L+2,4-D 2.137 mg/L;愈伤组织的继代周期为10~12 d。 Objective:To seek optimum combination of hormone concentration during callus subculture of Glycyrrhiza uralensis. Methods: The seed of Glycyrrhiza uralensis as explants, First generation callus induced by embryo root and hypocotyledonary axis of sterilized seed as material, The mathematical model were established by quadratic orthogonal rotary composite experimental design. The concentration of hormones were optimized. The callus growth dynamics of Glycyrrhiza uralensis were explored under optimized culture condition. Result: Coefficient of determination of model R2 is 0. 9161. The optimum combination of hormone concentration during callus subculture of Glycyrhiza uralensis is 6-BA 1. 514 mg/L, NAA 2. 059 mg/L, 2,4-D 2. 137 mg/L, the cycle of callus subculture is 10-12 days.
