摘要
目的探讨奥卡西平(OXC)对戊四氮(PTZ)致未成年大鼠认知功能的影响,为其在儿童患者中应用的安全性提供理论依据。方法利用PTZ建立致未成年大鼠模型,致大鼠共分为3组,分别予OXC、苯巴比妥(PB)和9g.L-1盐水灌胃,在实验的3个阶段通过Morris水迷宫实验评价各组大鼠认知功能的变化,并与未致大鼠进行比较(实验前进行情感运动能力检测以减少误差)。结果 1.各组大鼠情感运动检测结果差异无统计学意义。2.致大鼠空间定位能力受损,但这种受损经过训练后可恢复正常,服用OXC的致及正常大鼠空间定位能力均不受影响。3.致对大鼠学习进度的影响不明显,服用PB的致大鼠学习进度受损,服用OXC的致及正常大鼠学习进度均无明显受损表现。4.致可导致大鼠长时记忆能力一过性受损,服用PB的致大鼠受损更持久,服用OXC的致大鼠这种能力受到保护。5.致及服用PB和OXC对大鼠的短时记忆能力影响均不明显。结论癫不影响未成年大鼠的情感运动,但可导致其认知功能一过性受损,OXC对PTZ致未成年大鼠早期的认知功能损伤(主要体现在空间定位和长时记忆能力方面)有一定的改善作用;正常未成年大鼠服用OXC后的认知功能无明显受损表现。
Objective To discuss the effect of oxcarbazepine(OXC) on the cognitive function of the young rats with epilepsy(EP) induced by pentylenetetrazol(PTZ),then to provide the theoretical basis of its application safely to epileptic children.Methods The models of the epileptic young rats were established by PTZ,then the epileptic rats were divided into 3 groups and were given OXC,phenobarbital(PB) and 9 g.L-1 sodium chloride,respectively.In 3 phases of the experiment,they were appraised for the cognitive functions by Morris water maze test and compared with those of the normal rats(checking their sensibilities and movements before the test for getting rid of the errors).Results 1.The grades of sensibilities and movements of each group were not different statistically.2.The spatial orientation capabilities of epileptic rats were damaged,but the damage recovered after training,but the capabilities of epileptic and normal rats taking OXC were not affected.3.The effect of EP on the studying rate of progress was not obvious,the progress of epileptic rats taking PB was damaged,and the da-mage to epileptic and normal rats taking OXC was not obvious.4.EP could damage the long-term memory of the rats transitorily,the damage to the epileptic rats taking PB was durable,and the memory of epileptic rats taking OXC was protected.5.The effects of EP and taking PB or OXC on the capability of the short-term memory were not obvious.Conclusions EP does not affect the sensibilities and movements of the young rats,but it can affect their cognitive functions transitorily.OXC can alleviate the early cognitive function damage(especially on the capability of the spatial orientation and the long-term memory) of the epileptic young rats,and normal young rats do not have the obvious cognitive function damage after taking OXC.
