两步化学转化法构建猪圆环病毒2型ORF2基因重组腺病毒质粒 被引量:3

摘要 为了利用化学转化法快速构建携带猪圆环病毒2型ORF2基因的重组腺病毒质粒以便获得表达该基因的重组腺病毒,首先把骨架质粒pAdeasy-1通过化学法转化到氯化钙法制备的E.coliB J5183感受态菌中,通过含氨苄青霉素的LB平板筛选出含骨架质粒的阳性菌B J5183/p,再次用化学法把插入有猪圆环病毒2型ORF2基因的穿梭质粒rpShuttle-CMV-276经Pm eⅠ酶切线性化后转化到B J5183/p感受态细胞中进行同源重组,再在含卡那霉素的LB平板上培养,筛选出卡那霉素抗性菌进行质粒抽提纯化,酶切并电泳鉴定,获得重组阳性质粒rPAD-276。结果表明,运用2步化学转化法构建重组腺病毒质粒是一种简便、快捷且容易操作的方法。
