N,N'-二乙酰胱氨酸对半乳糖胺引起大鼠急性肝功能衰竭的治疗作用 被引量:1

Therapeutic effects of N,N'-diacetylcystine on acute liver failure induced by galactosamine in rats

摘要 目的观察N,N'-二乙酰胱氨酸(DAC)对半乳糖胺(Gal)引起的大鼠急性肝功能衰竭的治疗作用。方法 SD大鼠随机分为正常对照组,模型组,促肝细胞生长素(pHGF)组,DAC低、中、高剂量组,每组24只。除正常对照组外,每组腹腔注射Gal 1650 mg·kg^(-1),给药组于注入Gal前、后1 h分别腹腔注射pHGF 60 mg·kg^(-1),DAC 25、100、400 mg·kg^(-1),正常对照组和模型组腹腔注射等容量生理盐水。造模后30 h测定大鼠血清转氨酶、一氧化氮(NO)、丙二醛(MDA)和肝线粒体共轭二烯含量。结果与模型组比较,DAC能明显阻止Gal引起的大鼠血清转氨酶活力增高(P<0.01),降低血清MDA和NO水平(P<0.05),抑制肝线粒体共轭二烯含量(p<0.05),并减轻病理损害使大鼠死亡率显著降低(P<0.05)。结论 DAC对Gal引起的大鼠急性肝功能衰竭有显著的治疗作用。 AIM To observe the therapeutic effects of N,N'-diacetylcystine(DAC) on acute liver failure induced by galactosamine(Gal) in rats.METHODS SD rats were randomly divided into normal control group,model control group,positive control group and three DAC groups(n = 24,each group).Acute liver failure was induced by injection of Gal 1 650 mg·kg^(-1).One hour before and alter the injection of Gal,rats were administered intraperitoneally with saline(in the normal control group and model control group),hepatic growth factor(pHGF) 60 mg·kg^(-1)(in the positive control group),and DAC 25 mg·kg^(-1),100 mg·kg^(-1),400 mg·kg^(-1) (in three DAC group),respectively.RESULTS Compared with the model control group,DAC could prevent the increase of aminotransferase activity caused by Gal(P0.01),decrease MDA and NO level in serum(P0.05),inhibit conjugated diene level in liver mitochondrial(P0.05),decrease liver injury and mortality of rats(P0.05).CONCLUSION DAC is effective in the treatment of acute liver failure induced by Gal in rats.
