摘要
目的观察疏肝健脾方对乳腺增生大鼠血清sFas、乳腺组织Fas、FasL的影响。方法 60只大鼠随机分为空白对照组、疾病模型组、三苯氧胺组、疏肝健脾方小剂量组、疏肝健脾方大剂量组5组,每组12只,采用苯甲酸雌二醇和黄体酮肌注造模的同时,分别以蒸馏水、三苯氧胺、疏肝健脾方灌胃干预治疗30天后,观察乳腺增生大鼠血清sFas浓度和乳腺组织Fas、FasL的表达。结果 Fas在乳腺增生病模型大鼠中表达较低,FasL表达较高,经药物干预后,均能够增加Fas在乳腺组织中的表达(P<0.01),降低FasL中的表达(P<0.01),接近空白对照组,各组药物之间无显著性差异(P>0.05)。sFas在大鼠乳腺增生病模型血清中的浓度增高,经药物干预后能够降低sFas在大鼠血清中的浓度(P<0.01或0.05),各药物组之间无显著性差异(P>0.05)。结论疏肝健脾方对大鼠乳腺增生组织Fas/FasL系统有显著的干预作用,通过提高Fas降低FasL蛋白的表达,降低sFas在大鼠血清中的浓度,从而抑制/逆转乳腺增生,降低增生程度。
Objective To detect the effects of liver - smoothing and spleen - strengthening formula(LSF) on sFas and Fas/Fasl expression of hyperplasia histology of mammary gland(HMG) in rats. Methods 60 rats were divided into 5 groups at random with 12 rats in each group:blank control group, disease model group, tamoxifen group, LSF low dose group and LSH high dose group. Rats were injected with benzestrofol and progesterone to establish the HMG model. Meanwhile, every rat was administrated pure water, tamoxifen, low - dose LSF and high - dose LSF respectively once a day. At the 30th day, sFas concentration and Fas/Fsal expression level of HMG were observed. Result It was lower expression level of Fas and higher expression level of Fasl in the HMG model group. After those dugs were administrated respectively, there was increase in the expression level of Fas(P 〈 0.01 ) and decrease in the expression level of Fasl(P 〈 0.01 ), which was approximate to the blank control group. No significant difference existed among drug groups(P 〉 0.05 ). It was higher for sFas in HMG model group and after the intervention of the drugs, and sFas concentration was decreased (P 〈 0. 01 or P 〈 0. 05 ). Conclusion LSF can have significant intervention effects on Fas/Fasl system of HMG in rats, and cut down/reverse the hyperplasia of mam- mary gland by improving the expression level of Fas and degrade the expression level of Fasl and the concentration of sFas.
