25%蔗糖磷酸盐缓冲液对肝和肺组织核酸的保护作用

摘要 目的 探讨蔗糖磷酸缓冲液作标本保存剂对不同组织标本中核酸的保护作用.方法 用25%蔗糖磷酸缓冲液固定小鼠肝脏和肺组织2~72 h后,检测RNA完整性和纯度.结果 肝组织经25%蔗糖磷酸缓冲液保存48 h内,从中提取的RNA完整性与液氮保存的组织无明显区别,72 h内有明显差别;而肺组织保存72 h内无明显差别.结论 25%蔗糖磷酸缓冲液固定肝组织48 h内,不影响组织标本的RNA提取,而肺组织72 h内不影响组织标本的RNA提取.25%蔗糖磷酸盐缓冲液对不同组织的核酸保护作用时间不同. Objective To investigate the protection effect of 25 % sucrose in PBS on RNA from the different tissue samples. Methods Livers and lungs of mice were removed and incubated in 25% sucrose in PBS for 2 h - 72 h. Integrality and Purity of the tissues total RNA were detected after different treatments. Results No distinctive differences were observed between the RNA from incubated livers treated with 25% sucrose in PBS in 48h and that from liver samples in liquid nitrogen. No distinctive differences were observed between the RNA from incubated lungs treated with 25% sucrose in PBS in 72 h and that from lung samples in liquid nitrogen. Conclusion 25% sucrose in PBS did not effect the extraction of liver RNA in 48 h whle 25% sucrose in PBS did not effect the extraction of liver RNA in 72 h. The effect of 25% sucrose in PBS on RNA from the different tissue samples is different.
