摘要
目的:研究人骨髓间充质干细胞(hMSCs)在体外诱导分化为人汗腺细胞(hSGCs)。方法:体外分别分离培养hMSCs和hSGCs,原代培养hSGCs融合后,收集细胞,制备汗腺细胞匀浆,用含10%汗腺细胞匀浆的汗腺培养基来诱导培养hMSCs。1周后,检测诱导培养的hMSCs的抗原表达。结果:培养的hSGCs表达CK19和CEA;hMSCs表达CD44、105,不表达CK19、CD34和CEA。诱导培养1周,免疫细胞化学染色示诱导培养组少量细胞表达CEA和CK19,而对照组未见CEA和CK19阳性细胞;流式细胞仪检测CEA在诱导培养组的阳性表达率约为(6.2±1.2)%,对照组(0.9%±0.0)%(P<0.05)。结论:在无完整汗腺细胞存在的情况下,hMSCs在体外也可诱导分化为hSGCs。
Objective:To investigate the cellular phenotype conversion of human mesenchymal stem cells(hMSCs)induced into human sweat gland cells(hSGCs)by induced culture in vitro.Methods:HMSCs and hSGCs were isolated and cultured respectively.After the primary cultured hSGCs was confluence,they were collected and the cell homogenate was prepared.HMSCs were cultured with sweat gland cell medium supplemented with 10% sweat gland cell homogenate.One week later, the induced cells were detected for antigen expression.Results:Cultured hSGCs expressed CK19 and CEA,and hMSCs expressed CD44,105,but not CK19,CD34 or CEA.One week after the induced culture,the immunocytochemical staining showed a few induced cells expressed CEA or CK19,while there was no positive cells in the control group,and the flow cytometry showed the positive rate of CEA in the induced group was(6.2±1.2)%,while that in control group was(0.9±0.0)%.There was significant difference between two groups in the positive rate of CEA(P0.05).Conclusion:HMSCs can be induced into the phenotype of hSGCs in absence of intact sweat gland cells.
