摘要 [目的]研究枯草芽孢杆菌B26抑菌活性成分的稳定性。[方法]枯草芽孢杆菌B26发酵液离心过微孔滤膜后,用70%的(NH4)2SO2沉淀,透析后获得仍具有拮抗多隔镰孢霉的无菌粗提物,用平板涂布打孔法检测粗提物对温度、酸碱度、紫外线照射、有机溶剂和蛋白酶处理的稳定性。[结果]经100℃处理后,枯草芽孢杆菌B26粗提物抑菌直径是常温下的78.2%,但经121℃处理后,B26粗提物完全失去活性;在pH值为3~10范围内,枯草芽孢杆菌B26粗提物均具有抑菌活性,在pH值为7时其抑菌活性最大;枯草芽孢杆菌B26粗提物经距离为40cm、功率为20W的紫外灯照射80min后,其抑菌活性为对照的78.1%。经乙醚、甲醇、氯仿和丙酮处理30min后,枯草芽孢杆菌B26粗提物抑菌活性分别是对照的96.4%、85.7%、82.1%、81.5%。枯草芽孢杆菌B26粗提物经蛋白酶K、胰蛋白酶和胃蛋白酶处理后,其拮抗直径分别为15.2、16.2和16.3mm,抑制率分别是对照的76.7%、81.8%和83.3%。[结论]枯草芽孢杆菌B26粗提物抑菌具有较高的稳定性。 [Objective] The stability of antimicrobial active ingredients of Bacillus subtilis B26 was studied. [Method] The Fermentation broth of B. subtilis B26 was passed through porous membrane after centrifugation,70% (NH4)2SO2 was used to carry out precipitation,and the sterile crude extracts with resistance against Fusarium decemcellulare Brick were still obtained after dialysis. Plate coated perforated method was adopted to detect the treatment stability towards crude extracts on temperature,pH,ultraviolet radiation,organic solvents and protease. [Result] The inhibition diameter of crude extracts of B. subtilis B26 after treatment at 100 ℃ was 78.2% of the treatment at room temperature,but the crude extracts of B. subtilis B26 completely lost their activity after treatment at 121 ℃; the crude extracts of B. subtilis B26 all had antimicrobial activity in the pH value range of 3-10,and the antimicrobial activity was the largest when the pH value was 7.0; when the crude extract of B. subtilis B26 was irradiated by UV light (power 20 W) for 80 min from 40 cm distance away,the antimicrobial activity of which was 78.1% of the control; when it was treated with ether,chloroform,methanol and acetone for 30 min,the antimicrobial activities of crude extracts of B. subtilis B26 were 96.4%,85.7%,82.1% and 81.5% of the control,respectively; when it was treated with proteinase K,trypsin and pepsin,the inhibition diameters of crude extracts of B. subtilis B26 were 15.2,16.2 and 16.3 mm,the antimicrobial activities were 76.7%,81.8% and 83.3% of the control,respectively. [Conclusion] The crude extracts of B. subtilis B26 had higher antimicrobial stability.
