三氧化二砷诱导神经母细胞瘤细胞凋亡及对livin基因表达的影响

THE EFFECTS OF As_2O_3 ON THE APOPTOSIS OF HUMAN NEUROBLASTOMA CELL SK-H-SH AND ON THE EXPRESSIONS OF LIVIN GENE

目的探讨三氧化二砷(As2O3)诱导人神经母细胞瘤SK-N-SH细胞凋亡效应及其对livin基因表达的影响。方法比色法观察As2O3对神经母细胞瘤SK-N-SH细胞增殖的影响;Annexin V荧光探针和流式细胞仪检测早期凋亡细胞;RT-PCR分析转染前后livin mRNA的表达;Western检测Livin蛋白的表达。结果 As2O3明显抑制SK-N-SH细胞的增殖;As2O3诱导SK-N-SH细胞凋亡时livin基因表达降低。结论 livin基因表达降低可能是As2O3诱导神经母细胞瘤细胞凋亡的机制之一。

Objective To investigate the effect of As2O3 on the apoptosis of human neuroblastoma cell SK-H-SH and on the expressions of livin gene.Methods The effect of As2O3 on the proliferation of neuroblastoma cells was measured with colorimetry.Annexin V fluorescein probes and flow cytometry were used to detect the early apoptotic cells.The expression level of livin mRNA was assayed by RT-PCR,and the expressions of Livin protein were detected by western blot.Results As2O3 significantly inhibited the proliferation of SK-N-SH cells.The expressions of livin gene decreased while inducing the apoptosis.Conclusion The decrease in the expressions of the livin gene may be one of the mechanisms through which As2O3 induces the apoptosis of SK-N-SH cells.
