应用猪瘟抗原/血清ELISA测定猪瘟兔化弱毒疫苗中的病毒含量 被引量:8

摘要 简单、快速地测定猪瘟病毒的含量是猪瘟兔化弱毒疫苗(HCLV)生产工艺中难以解决的技术之一。本试验通过应用猪瘟抗原/血清ELISA试剂盒(IDEXX HerdCheck Co.),对已知滴度的24份HCLV半成品细胞苗和21份成品疫苗进行抗原含量测定,结果24份HCLV半成品细胞苗95.83%与兔体定型热反应结果相符,检测敏感性可达20头份/瓶(或病毒拷贝数109个/瓶)。但该方法无法区分猪瘟病毒抗原与牛流行性腹泻病毒抗原。试验结果证明,IDEXX猪瘟抗原/血清ELISA试剂盒能够稳定、快速、简单地用于HCLV细胞苗的半成品与成品的病毒含量测定,为疫苗生产的质量控制提供了一种可靠和稳定的方法。 Detecting virus content in hog cholera lapinized vaccine simply and rapidly is one of the technologies difficult to solve in the HCLV production process.In this experiment,the classical swine fever antigen/serum ELISA kit(IDEXX Herd Check Co.) is used to detect the virus content of 24 batches of semi-finished HCLV and 21 batches of finished HCLV.The results indicate that the high accordance rate between rabbit heat reaction and ELISA detection in 24 batches of semi-finished HCLV reaches 95.83%.In addition,the sensitivity of the antigen/serum ELISA kit runs to 20 dosages every bottle(or 109 copies every bottle).However,this method can not distinguish the classical swine fever virus antigen from Bovine viral diarrhea virus antigen.As a result,the classical swine fever antigen/serum ELISA kit(IDEXX Herd Check Co.) can be used in detecting the virus content of semi-finished and finished HCLV stably,simply and rapidly,and it is suitable for quality control in the HCLV production process.
