阿魏侧耳多糖抗癌作用机理研究 被引量:11

摘要 目的:阿魏侧耳(阿魏菇)多糖(Pleurotus ferulae polysaccharide,PFP)抗宫颈癌作用的机理研究。方法:MTT法、中性红比色法和Griess法。结果:灌胃给药500 mg.kg-1的阿魏菇多糖可抑制小鼠宫颈癌U14移植瘤的生长,抑瘤率达到39%,临床抗宫颈癌药顺铂的抑瘤率为46.5%,阿魏菇多糖与顺铂共同作用时,抑瘤率达到65.1%;但是在体外实验中,不同浓度阿魏菇多糖分别作用于人宫颈癌HeLa和人乳腺癌MCF-7细胞不同时间后,却不能抑制两种细胞的生长;在5μg.mL-1的脂多糖刺激下,阿魏菇多糖不能促进脾淋巴细胞的增殖;不同浓度的阿魏菇多糖作用于腹腔巨噬细胞24 h,能使腹腔巨噬细胞的吞噬能力显著增强,并且呈现明显的剂量依赖性,与此同时阿魏菇多糖也促进了腹腔巨噬细胞的增殖,但是却不能刺激腹腔巨噬细胞释放NO。结论:阿魏菇的抗宫颈癌机制可能是通过促进小鼠机体免疫功能而实现的。 Objective: To investigate the mechanisms of anti-tumor effect of Pleurotus ferulae polysaccharide (PFP) on cervix. Methods: MTT assay, neutral red colorimetry and Griess assay were used. Results: 500mg· kg^-1 PFP (i. g. ) can restrain growth of transplantation tumor U14 of cancer of the cervix(CACX)in Kunming mice and the inhibition rate was 39 %. In combination PFP with Diaminodichloroplatin, the inhibition rate against CACX U14 was 65. 1%, compared with the inhibition rate of 46.5 % in Diaminodichloroplatin-treated alone group. However, after treated HeLa and MCF-7 with different concentrations of PFP for different time periods, respectively, the PFP can not inhibit cell growth of the two kinds, in vitro. In the presence of 5 ~xg ~ mLl LPS, the PFP can not induce proliferation of splenic lymphocyte. Wherever, when the peritoneal macrophages were cultured with different concentrations of PFP for 24 h, the PFP was found to significantly increase the phagocytosis in a concentration- dependent manner and trigger the proliferation of peritoneal macrophages, however, NO could not be detected in the same peritoneal macrophages. Conclusion: The PFP inhibited the CACX growth by means of improving the immune system in Kunming mice.
