FAS和E-FABP在糖尿病大鼠视网膜中的表达及意义 被引量:1

摘要 目的探讨脂肪酸合成酶(FAS)和表皮型脂肪酸结合蛋白(E-FABP)与糖尿病视网膜病变的关系及作用机制。方法用半定量逆转录聚合酶链反应、免疫组织化学方法检测FAS和E-FABP在Alloxan诱导糖尿病大鼠视网膜中的表达变化。结果 FAS主要于糖尿病大鼠视网膜视锥视杆细胞内节段表达明显上调,mRNA表达量比正常对照组明显升高(P<0.01)。而E-FABP于外核层、外丛状层、内核层、内丛状层及神经节细胞层表达,和对照组相比表达量无明显差异,mRNA和蛋白的表达量无明显变化(P<0.05)。结论 FAS可能通过异常的脂代谢途径在糖尿病视网膜病变过程中其重要的作用。E-FABP在细胞内外脂肪酸的共同调节作用下表达无明显差异。 Objective To detect the role of fatty acid synthase(FAS) and epidermal fatty acid binding protein(E-FABP) in the diabetic retinopathy pathogenesis.Methods The expressions FAS and E-FABP in the retina of alloxan induced diabetic SD rats were detected by RT-PCR and immunohistochemistry analysis.Results The retinal FAS mRNA expression abundance of the 12 weeks diabetic rats were remarkable increased compared with that of the control rats(P0.01).The mRNA expression abundance of E-FABP of the two groups was no different.Through the immunohistochemcal staining,the FAS positive stains were mainly observed in the cone and rod inner segment.E-FABP positive stains were mainly observed in ONL,OPL,INL,IPL and GCL.There was no significant difference between experimental group and control group.Conclusion The increasing expression of FAS in the retina of diabetic model rats suggest that FAS maybe plays an important role in the genesis and development of diabetic retinopathy through abnormal adipose metabolism pathway.
