摘要
目的研究5-Aza-CdR与低浓度CDDP联合用药对胃癌细胞系生长及DAPK1的影响。方法选择CDDP(1×10-6 mol/L)与1×10-6 mol/L的5-Aza-CdR分别处理体外培养的BGC823细胞与SCG7901细胞,5-Aza-CdR与CDDP联合处理体外培养的细胞后,用MTT法检测药物处理后的细胞增殖活性;RT-PCR法检测用药前后细胞中DAPK1的mRNA表达的变化。结果与单独用药组比较,胃癌细胞在药物联合作用组生长增殖活性明显被抑制。联合用药组DAPK-1基因的mRNA表达明显增加。结论 5-Aza-CdR与CDDP联用对胃癌细胞的生长抑制具有协同作用。
Objective To observe the effects of 5-Aza-CdR and low concentration CDDP on proliferation of human gastric cancer cell lines and the expression of DAPK-1 gene. Methods BGC823 and SCG7901 cells were cultured in RPMI1640,and they were treated with 5-Aza-CdR(1×10-6 mol/L),CDDP(1×10-6 mol/L),and both 5-Aza-CdR(1×10-6 mol/L)and CDDP(1×10-6 mol/L).The proliferation of the cells was detected by MTT assay.The expression of DAPK1 was detected by RT-PCR. Results Cells were exposed to 5-Aza-CdR(1×10-6 mol/L)and CDDP(1×10-6 mol/L),the growth of cells was inhibited more obviously.The expression of DAPK1 mRNA obviously increased in 5-Aza-CdR(1×10-6 mol/L)and CDDP(1×10-6 mol/L). Conclusion The gastric cancer cells growth treated with both 5-Aza-CdR and CDDP are inhibited more obviously.
