摘要
目前提取总RNA普遍使用的是酸苯酚萃取法(或称硫氰酸胍法).此法所提取的RNA中存有少量基因组DNA的污染.为了克服这一缺点,我们对酸苯酚提取RNA的方法进行了改进.即先用酸苯酚萃取,然后用蛋白酶K消化,再次酸苯酚萃取.
A method for isolating high pruified total RNA from a sample of flesh or frozen tissues and cells is described. The results of the common acid phenol extraction method show that the RNA isolate was contaminated by a small amount of genomic DNA. In order to overcome this shortcoming, an improved procedure is used by digesting the isolate, which derived from the first acid phenol extraction, with proteinase K and then following an acid phenol extraction once more.
