酸苯酚萃取总RNA方法的改进

THE IMPROVEMENT OF ACID PHENOL EXTRACTION METHOD OF ISOLATING TOTAL RNA

目前提取总ＲＮＡ普遍使用的是酸苯酚萃取法（或称硫氰酸胍法）．此法所提取的ＲＮＡ中存有少量基因组ＤＮＡ的污染．为了克服这一缺点，我们对酸苯酚提取ＲＮＡ的方法进行了改进．即先用酸苯酚萃取，然后用蛋白酶Ｋ消化，再次酸苯酚萃取．

A method for isolating high pruified total RNA from a sample of flesh or frozen tissues and cells is described. The results of the common acid phenol extraction method show that the RNA isolate was contaminated by a small amount of genomic DNA. In order to overcome this shortcoming, an improved procedure is used by digesting the isolate, which derived from the first acid phenol extraction, with proteinase K and then following an acid phenol extraction once more.
