摘要
目的选育抗真菌抗生素梧宁霉素高产菌株。方法本文通过紫外线诱变的方式对不吸水链霉菌梧州亚种04(Streptomyces ahygroscopicus subsp.Wuzhouensis 04)进行菌种选育,并使用白念珠菌作为生物活性鉴定菌筛选高产突变株。结果经筛选得到一株具有很高抑菌活性的突变株,在发酵液表观粘度、菌丝形态、组分分布等方面与出发菌株有明显差异。结论经高效液相色谱及飞行时间质谱对产物的分析,证实该突变株为原出发菌株中含量较低的茴香霉素的高产菌株。
Objective To breed antifungal antibiotic wuningmeisu C high yield strain. Methods Streptomyces ahygroscopicus subsp, wuzhouensis 04 was mutated by UV mutagenesis. The bioactivity of high yield mutants was tested by the biological assay using Candida Albicans. Results A high yield strain was obtained by screening which was different from the original strain on the facet of apparent viscosity of the fermentation broth, the hyphal and the component distribution etc. Conclusion The analysis by high performance liquid chromatography and time of flight mass spectrometry showed that the yield of Wuningmeisu C (anisomycin) was very low in the original strain, whereas high in this mutant.
