摘要
建立了用高效液相色谱—电化学检测器((HPLC/ECD)、柱前衍生法,同时测定生物样本中组织胺(Hist)和去甲肾上腺素(NA)的方法.可在20min内分离检测出Hist和NA,线性范围为0.015~50mg/L,最低检出限为15ng/L,回收率为95%,精密度为5.8%~11.1%.具有灵敏、稳定、特异性高等特点.应用检测了70例健康人、5例高血压病患者血浆中Hist和NA的含量。
A method of simultaneous determination of Histamine (Hist) and Noradrenaline (NA) in human
plasma was established to study the decomposing relationship of Hist and NA. The assay was
based on precolumn derivatization of Hist and NA ,internal standard with 3 methylhistamine (3
Me hist), separation of the derivatives by high performance liquid chromatographic method with
electrochemical detection (HPLC/ECD ).This complete separation of 3 kinds of component was
achieved within 20 min .The assay was linear from 0.015~50ng/ml.The minimal detection limit
was 15ng/L.The recovery of 3 kinds of component added to plasma samples was 95%.The
precision was 5.8%~11.1%. HPLC/ECD method has many advantages in rapidity, simplicity,
accuracy, sensitivity and higher distinction. The experimental results have indicated that the
distribution of Hist parallels with that of NA in human plasma.
