不同经典方剂对肝外胆汁淤积大鼠肝肾功能的影响

Effect of Different Classic Formulas on the Liver and Renal Function of Extrahepatic Cholestasis in Rats

目的比较茵陈蒿汤、茵陈五苓散、大柴胡汤对实验性肝外胆汁淤积大鼠肝肾功能的影响。方法行胆总管三段结扎术复制大鼠肝外胆汁淤积模型,造模1周后各药物干预组每天分别灌胃给予1ml/100g体重茵陈蒿汤、茵陈五苓散和大柴胡汤,模型组和假手术组给予等体积生理盐水。至造模3周时检测肝肾功能。结果与假手术组比较,模型组血清白蛋白(ALB)含量显著降低(P<0.05),总胆红素(TBiL)、总胆汁酸(TBA)、胆固醇(CHO)、尿素氮(BUN)含量,丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)、碱性磷酸酶(ALP)、γ-谷氨酰转肽酶(GGT)活性显著增高(P<0.05或P<0.01);与模型组比较,茵陈蒿汤、茵陈五苓散和大柴胡汤均可显著增加白蛋白(ALB)含量;显著降低TBiL、TBA含量,ALT、AST、LDH、ALP、GGT活性(P<0.01);茵陈蒿汤在降低TBiL含量、ALP活性方面显著优于茵陈五苓散组(P<0.01);茵陈蒿汤在降低ALT、AST、LDH、GGT活性,TBA含量方面显著优于大柴胡汤组(P<0.01)。结论茵陈蒿汤、茵陈五苓散和大柴胡汤均能有效改善BDL所诱发肝外胆汁淤积大鼠模型肝肾功能改变,茵陈蒿汤综合疗效较优。

Objective To compare the effect of the three classic formulas Yinchenhao Decoction（Decoction of Herba Artemisiae Scopariae）,Yichen Wuling Powder（Powder of Five Drugs with Poria added by Herba Artemisiae Scopariae）and Dachaihu Decoction（Major Bupleurum Decoction）on the liver and renal functions of rats with experimental extrahepatic cholestasis.Methods Common bile duct ligation in three segments was performed to establish the cholestasis models.One week after modeling,the intervention groups were respectively given Yinchenhao Decoction,Yinchen Wuling Powder and Dachaihu Decoction intragastrically everyday in dose of 1ml/100g body weight.The model group and sham operation groups were administered isometric normal saline.Rats were killed 3 weeks after modeling to detect the liver and renal functions.Results Compared with the sham operation group,ALB content in the model group was significantly decreased（P0.05）,the content of TBiL,TBA,CHO,BUN,and the activity of ALT,AST,LDH,ALP and GGT were all significantly increased（P0.05,P0.01）.Compared with the model group,Yinchenhao Decoction,Yinchen Wuling Powder,and Dachaihu Decoction all could increase the content of ALB,and significantly decrease the content of TBiL and TBA,and the activity of ALT,AST,LDH,ALP and GGT（P0.01）.Yinchenhao Decoction was significantly better than Yinchen Wuling Powder in decreasing TBiL content and ALP activity（P0.05）.Yinchenhao Decoction was significantly better than Dachaihu Decoction in decreasing the activity of ALT,AST,LDH and GGT,and the content of TBA（P0.01）.Conclusion Yinchenhao Decoction,Yinchen Wuling Powder and Dachaihu Decoction all can improve the changes of liver and renal functions of rats with extrahepatic choleasstis.The comprehensive effect of Yinchenhao Decoction is better.
