CD_(44)与nm23在神经母细胞瘤中的表达及意义

摘要 目的检测CD44与nm23基因在神经母细胞瘤(NB)中的表达,探讨CD44和nm23 mRNA的表达与NB发生及转移的关系。方法选择30例术前未经化疗的NB患儿为研究对象,收集术中切除的无坏死新鲜瘤体组织为研究组,相应瘤旁组织为对照组,将标本放置于-80℃冰箱保存。依据Trizol试剂说明书于超净工作台分别提取NB和瘤旁组织mRNA,将其反转录为cDNA后进行扩增内参和引物,凝胶电泳检测nm23 mRNA及CD44 mRNA在NB和瘤旁组织中的表达水平。结果 nm23 mRNA在研究组及对照组中均有表达,但研究组表达低于对照组,研究组中有淋巴结转移组织中的表达低于无淋巴结转移组织中的表达。CD44 mRNA亦在研究组和对照组中均有表达,但研究组表达显著高于对照组,且研究组有淋巴结转移组织中的表达高于在无淋巴结转移组织中的表达。结论 nm23 mRNA及CD44 mRNA的表达水平与NB发生、发展和肿瘤转移密切相关,nm23 mRNA具有抑制NB发生及转移功能,而CD44 mRNA则促进NB的发生及转移。采用PCR技术可以有效检测NB nm23 mRNA及CD44 mRNA的表达水平,有助于判断其生物学行为。 Objective To detect the gene expressions of nm23 and CD44 in neuroblastoma(NB) and investigate their correlations with formation and metastatic of NB.Methods Thirty normal tissues near the tumor(control group) and primary cancer tisssues(experimental group)from 30 NB children were treated from Department of Pediatric Surgery.All the tissues were kept in-80 ℃ freezer instantly after operation.Total mRNA were extracted from specimens according to Trizol reagent protocol.After that mRNA were progressed directed to cDNA by revers transcription,then amplify β-actin and primer.The expressions of amplification production of nm23 mRNA and CD44 mRNA were detected by gel electrophoresis.Results The expression levels of nm23 mRNA and CD44 mRNA exist both of the experimental group and control group,but the expression level of nm23 mRNA in experimental group was lower than that in the control group.In experimental group,the expression level of nm23 mRNA in the tissues with lymph metastasis was lower than that in no lymph metastasis.The expression level of CD44 mRNA in experimental group was significantly higher than that in control group.In experimental group,the expression level of CD44 mRNA in the tissues with lymph metastasis was higher than that in no lymph metastasis.Conclusions The gene expressions of nm23 and CD44 are correlated with formation and metastasis of NB.nm23 mRNA inhibits the formation and metastatic of NB and CD44 mRNA promote formation and metastatic of NB.Polymerase chain reaction can effectively detects the gene expression levels of nm23 mRNA and CD44 mRNA.Examination of expression these 2 genes may be helpful to judge biological behavior of NB.
