摘要
目的:建立同时测定旋覆花配方颗粒中绿原酸及咖啡酸含量的HPLC方法。方法:采用正交试验优化旋覆花配方颗粒的提取工艺,用HPLC法测定旋覆花配方颗粒中绿原酸及咖啡酸的含量。色谱柱为Shim-packVP-ODS柱(250 nm×4.6 mm,5μm);流动相为乙腈-0.1%磷酸(9∶91);检测波长为327 nm;流速为1.0 mL.min-1。结果:绿原酸在0.86-2.58μg(r=0.999 9)范围内线性关系良好,平均回收率为102.95%,RSD为1.21%;咖啡酸在0.28-1.10μg(r=0.999 9)范围内线性关系良好,平均回收率为99.72%,RSD为1.64%。结论:该方法简便、快速、重复性好,可作为旋覆花配方颗粒中绿原酸及咖啡酸的含量测定方法。
Objective:To develop a HPLC method for the simultaneous determination of chlorogenic acid and caffeic acid in Xuanfuhua dispensing granule.Methods:The orthogonal test was used to optimize extract conditions.Chlorogenic acid and caffeic acid were separated on Shim-pack VP-ODS(250 mm×4.6 mm,5 μm) column and detected at 327 nm,with Acetonitrile-0.1% phosphoric acid solution(9∶91) as mobile phase.The flow rate was 1.0 mL·min-1.Results:The method had good linear relationship in the range of 0.86~2.58 μg(r=0.999 9) for chlorogenic acid and 0.28~1.10 μg(r=0.999 9) for caffeic acid,respectively.The results of the average recovery were 102.95% with RSD of 1.21% for chlorogenic acid and 99.72% with RSD of 1.64% for caffeic acid.Conclusion:The method was simple and reliable which can be used to simultaneously determine chlorogenic acid and caffeic acid in Xuanfuhua dispensing granule.
