来氟米特对糖尿病模型大鼠肾组织中血小板衍化生长因子的影响研究

摘要 目的:探讨来氟米特对糖尿病模型大鼠肾组织中血小板衍化生长因子(PDGF)的影响。方法:取大鼠单剂量注射链脲佐菌素60mg·kg-1建立糖尿病模型,建模成功后分为模型组(自来水)和治疗组(来氟米特,5mg·kg-1·d-1),每组16只,灌胃给予相应药物,于第4、8周末各组随机取样8只,检测血糖、胆固醇、甘油三酯、血肌酐、尿白蛋白、尿β2-微球蛋白(β2-MG)等生化指标及肾脏肥大指标的变化,并以免疫组化法检测肾组织中PDGF表达。另设正常对照组进行比较。结果:与模型组比较,治疗组大鼠第4周除血糖外、第8周除甘油三酯外,其他生化指标、肾脏肥大指标以及肾组织中PDGF表达均降低。结论:来氟米特可能通过下调糖尿病模型大鼠肾组织中PDGF表达来实现肾脏保护作用。 OBJECTIVE: To explore the effect of Leflunmide on platelet-derived growth factor (PDGF) of kidney tissue in diabetic rats. METHODS: Diabetic model was established by single dose of streptozotocin 60 mg.kg-1. Model rats were randomly divided into model group (tap water) and treatment group (Lefltmmide 5 mg.kg-l.d-1) with each group of 16 rats. The rats were sacrificed after 4 or 8 weeks of treatment. Plasma glucose, cholesterol, triglyceride, creatinine, urinay albumin, urinaryfβ2-MG and kidney hypertrophy index were measured. PDGF protein expression was measured by immunohistological staining and compared with normal control group. RESULTS: Compared with model group, the biochemical parameters, kidney hypertrophy index and PDGF protein expression in treatment group decreased except for blood glucose at 4th week and triglyceride at 8th week. CONCLU- SION: Leflmunide can reduce the expressions of PDGF in kidney of diabetic rats to protect kidney.
