摘要
目的:考察羟基喜树碱(HCPT)缓释片瘤内植入对C6脑胶质瘤模型大鼠的肿瘤抑制作用。方法:通过将C6脑胶质瘤细胞悬浮液接种于大鼠脑内,10d后建立C6脑胶质瘤模型,取模型大鼠分为模型对照组、空白对照组(植入不含HCPT的缓释片)、静脉注射治疗组(HCPT注射液,2mg·kg-1)、瘤内注射治疗组(HCPT注射液,约含HCPT2.4mg)、瘤内植入治疗组(HCPT缓释片,约含HCPT2.4mg),每组10只,给予相应药物,比较平均存活期,并用核磁共振成像(MRI)观测大鼠脑内肿瘤生长情况。结果:与模型对照组比较,除瘤内植入治疗组大鼠平均存活期明显延长(P<0.001)外,其余各组均无明显差异;MRI观测结果显示瘤内植入治疗组大鼠治疗后15d内肿瘤体积没有明显增大,且第15天略有缩小,其余各组肿瘤体积均增大。结论:HCPT缓释片瘤内植入明显延长了模型大鼠的平均存活期,但其本身对于脑组织的神经毒性作用还有待于考察。
OBJECTIVE: To investigate the inhibition effect of Hydroxycamptothecin sustained-released tablets (HCPT)implant- ed into brain on C6 glioma in rats. METHODS: The C6 glioma cells were transplanted into brain of rats, and C6 glioma model was established after 10 days. Model rats were divided into model control group, blank control group (implanting sustained-release tablets which were not containing HCPT), intravenous injection therapy group (HCPT injection 2 mg. kg ^-1), intratumoral injection therapy group (HCPT injection containing about HCPT 2.4 mg), carcinoma implantation therapy group (HCPT sustained-release tablets containing about 2.4 mg) with each group of 10 rats. The median survival time was compared among those groups after giving relevant drugs. The growth of glioma was detected by MRI dynamically. RESULTS: Compared with model control group, the median survival time of cerebral implantation group increased significantly while other groups had no significant difference. MRI results showed that tumor volume of cerebral implantation group had no significant increase after 15 days of treatment, and at 15th day tumor volume decreased slightly. The tumor volume of other groups all increased. CONCLUSION: HCPT sustained-release tablets implanted into brain can increase the median survival time of model rats significantly, and neurotoxic effect of it on cerebral tissue is to be proven by further experiment.
