摘要
目的:建立HPLC法测定三岛柴胡各部位的柴胡皂苷a、c、d的含量。方法:采用Symmetry C18(4.6 mm×150 mm,5μm)色谱柱,流动相为乙腈-水,梯度洗脱,流速1.0 mL.min-1,检测波长200 nm,柱温25℃。结果:1年生三岛柴胡中各部位柴胡皂苷a、c、d的含量均比2年生三岛柴胡的含量高。在1年生样品中种子含有的有效成分最多,根次之,茎最少;2年生样品中根含有的有效成分最多,种子次之,茎最少。结论:该方法为三岛柴胡不同药用部位的质量控制提供实验依据,该方法具有简便、快速、准确、重复性较好的特点。
Objective:To establish an HPLC for determination of saikosaponins a,c,d in different parts of Bupleurum falcatum.Methods:Symmetry C18(4.6 mm×150 mm,5 μm) column was adopted;The mobile phase was acetonitrile-water with gradient elution at the flow rate of 1.0 mL·min-1;The detection wavelength was 200 nm,and the column temperature was 25 ℃.Results:The contents of saikosaponins a,c,d in different parts of annual B.falcatum were higher than that of biennial B.falcatum,and the contents of three saikosaponins in the seeds of annual level B.falcatum were the highest,then followed by root and stem.Conclusion:The quantitative method for determination of saikosaponins a,c,d in different parts of B.falcatum by HPLC provided the basis for quality control of B.falcatum,this method is simple,rapid and accurate with good reproducibility.
