实时荧光定量PCR分析细菌的组成及数量 被引量:3

摘要 目的采用实时荧光定量PCR的方法对酸奶中的乳酸杆菌和双歧杆菌进行检测。方法通过标准菌株的培养和标准曲线的构建,建立检测乳酸杆菌和双歧杆菌的实时荧光定量PCR的检测方法,对送检的4组样品进行检测。结果通过标准曲线的构建得到乳酸杆菌和双歧杆菌的标准曲线方程:乳酸杆菌:Y=-3.293x+14.52;其r值为0.999;双歧杆菌为:Y=-3.464x+17.42,其r值为0.998,不同品牌的酸奶中,乳酸杆菌和双歧杆菌的数量存在很大差异,由于保密原因,本文将酸奶品牌简称为1、2、3、4,1和2两个品牌的酸奶中乳酸杆菌和双歧杆菌的数量均高于3和4号两个品牌,差异有统计学意义(P<0.05)。结论实时荧光定量PCR可以准确的对样品中乳酸杆菌和双歧杆菌的数量进行检测。
