摘要
目的 研究克罗卡林对心肌梗死面积及梗死部位心肌细胞内三磷酸腺苷 (ATP)含量的影响 ,并探讨其可能机制。方法 将 84只开胸安置缺血再灌注装置的Wistar大鼠分为四组 :对照组 ,缺血再灌注组 ,克罗卡林 +缺血再灌注组和克罗卡林 +优降糖 +缺血再灌注组 ,分别测定各组的血浆肌酸激酶浓度、梗死面积和梗死部位心肌细胞ATP含量。结果 缺血后血浆肌酸激酶含量升高 ,局部ATP浓度下降 ;克罗卡林能缩小心梗面积、降低血浆肌酸激酶浓度、减少梗死部位心肌细胞ATP的消耗 (P <0 .0 1) ;优降糖明显减弱克罗卡林的上述作用 (P <0 .0 1)。结论 克罗卡林对缺血心肌有明显的保护作用 ;
Objective To study the effect of cromakalim on infarct size and the amount of ATP on rat myocardium exposed to ischemia/reperfusion(I/R). Methods The snare for occlusion and reperfusion had been put into the main coronary artery in 84 Wistar rats which were divided into 4 groups: control, I/R, cromakalim+I/R and cromakalim+glibenclamide+I/R. The plasma concertation of creatine kinase, the infarct size and the myocardial level of ATP were measured in each group. Results The protective effect of cromakalim was evident: the infarct size, the concertation of creatine kinase were lower while the level of ATP was much higher than those of I/R group( P <0.01). Gliberclamide reduced the effect of cromakalim ( P <0.01). Conclusion Cromakalim protect the myocardium against ischemia/reperfusion damage, which the possible mechanism is activating the ATP sensitive potassium channel.
