卡马西平一次性给药方案维持夜间有效浓度的实验研究

Carbamazepine once daily in prevent of seizures at night or early morning

目的：考察顿服卡马西平治疗深夜或晨醒时发作的癫痫患者是否合理安全。方法：１８例癫痫患者随机分为两组，一组傍晚顿服卡马西平３００ｍｇ，另一组顿服２００ｍｇ。于服药后１２ｈ内多次取样，采用ＨＰＬＣ法测定其血药浓度。所得数据按血管外给药一房室数据进行拟合，算得其维持治疗浓度范围需要时间。结果：３００ｍｇ剂量组维持治疗浓度范围时间是７ｈ，２００ｍｇ剂量组维持治疗浓度范围时间是２ｈ，两者相比，有显著性差异。结论：顿服３００ｍｇ卡马西平治疗夜间或凌晨发作癫痫，能够维持７ｈ血药浓度在治疗范围内，符合给药目的。

OBJECTIVE:To study the possibility and safety when carbamazepin was given qn.to control seizures at night or early morning.METHOD:18 epilepsy patients were divided into two groups randomly.The patients were given carbamazepin 300mg qn.in one group or 200mg qn.in another one.Blood samples were obtained in the next 12 hours.The blood samples were analyzed by HPLC.The pharmacokinetic parameters were got with 3P87 program.RESULTS:When 300mg carbamazepin was given qn.,the Blood concentrations were kept about 7 hours in therapeutic range.But for the 200mg group,the blood concentrations maintained only 2 hours in therapeutic range.There was a signficant,difference between the two groups.CONCLUSION:Taking 300mg carbamazepin qn.may be suitable for the epilepsy patients whose seizures generally attacked at night or early morning.
