摘要
目的:探讨儿茶素诱导体外培养的LoVo大肠癌细胞株凋亡及放线菌酮和硫酸锌对儿茶素诱导细胞凋亡的拮抗作用。方法:用儿茶素处理LoVo细胞,加或不加放线菌酮和硫酸锌,用琼脂糖凝胶电泳、荧光显微镜和流式细胞仪方法,观察LoVo细胞生化和形态学方面的改变。结果:荧光显微镜下可见LoVo细胞在形态学上出现典型的细胞核固缩、碎裂等凋亡细胞的形态学改变;琼脂糖凝胶电泳呈典型的“梯状”(DNAladder)带,在同时加于放线菌酮或硫酸锌时,“梯状”带消失;PI染色的流式细胞仪直方图上出现亚二倍体峰,并具有明显的时间和剂量效应关系。结论:儿茶素能诱导大肠癌LoVo细胞的凋亡,这种凋亡作用能被放线菌酮或硫酸锌所拮抗。
Objective:
To investigate the apoptosis and its antagonism is induced by catechines in a colon cell
line,LoVo,in culture. Methods: After treatment of LoVo cells with catethines,in the presence or
the absence of cycloheximide and zinc sulfate,the apoptosis was detected by gel
electrophoresis,fluorescent light microscope and flow cytometry. Results: After the LoVo cells
were treated by catechines,it was shown that:1).Typical nuclear condensation and fragmentation
were observed by fluorescent light microscop.2).DNA ladderappeared on a 2%agarose
gel.However,the DNA ladder disappeared in the presence of cycloheximide or zinc
sulfate.3).Hypodiploid peak appeared on the histogram of flow cytometry with PI
staining.Moreover,there are concentrationdependent and timedependent effects on the
apoptosis of LoVo cells induced by catechines. Conclusion: Catechines have an effect of
inducing apoptosis of LoVo cells,which is antagonized by cycloheximide and zinc sulfate.
