摘要
目的:观察低强度中频超声对人卵巢癌细胞的杀伤作用。方法:取对数生长期的HO8910细胞,分成实验组和假辐照组,以低强度中频超声(频率1.7MHz)辐照。用MTT法计算细胞抑制率;光镜观察细胞形态,Hoechst33258荧光染色法检测凋亡细胞。结果:低强度中频超声照射各组间细胞抑制率与对照组相比有统计学差异,光镜下可见细胞形态学改变,同时Hoechst33258荧光染色法观察到亮蓝凝聚的凋亡细胞。结论:低强度中频超声对人卵巢癌HO8910细胞体外生长有明显杀伤作用,并可诱导其凋亡。
Objectives: The present study aims to investigate the cytatoxicity of low - intensity ultrasound on ovarian cancer cells. Metheds: H08910 cells were randomly divided into sham radiation group and experimental group. The cells in the experimental group were exposed to ultrasound wave ( 1.7MHz). The cytotoxicity was measured by MTF assay. The morphological changes of HO8910 cells were observed under light microscope. Apoptosis was analyzed using fluorescent microscopy with Hcechst 33258 stainin. Results: The cytotoxicity was significant difference between experimental group and sham radiation group. Light microscope confirmed the above results, Apoptosis were found by Hoechst 33258 staining method. Conclusion: Low- intensity ultrasound dan significantly kill ovarian cancer cell and induce apoptosis in vitro.
