摘要
目的观察炎症状态下,大鼠大网膜Ⅰ型乳斑面积、数量及巨噬细胞超微结构的变化,探讨Ⅰ型乳斑在腹腔中的作用。方法 SD大鼠腹腔注射沙培林诱导免疫反应,微粒子活性炭和HE染色双重标记大网膜乳斑,Im age-Pro P lus软件测定Ⅰ型乳斑的数量和面积,电镜观察两组Ⅰ型乳斑内巨噬细胞的超微结构。结果实验组与对照组相比,Ⅰ型乳斑面积显著增大[(0.316±0.066)mm2比(0.102±0.041)mm2,P<0.01],Ⅰ型乳斑数量显著增多[(11.053±1.543)个/cm2比(1.913±0.851)个/cm2,P<0.01]。实验组Ⅰ型乳斑巨噬细胞内的线粒体、溶酶体、高尔基体等细胞器丰富,粗面内质网肿胀扩张,伪足粗大密集。结论沙培林使Ⅰ型乳斑数量增多、面积增大,并能够激活Ⅰ型乳斑内的巨噬细胞,表明Ⅰ型乳斑在腹腔内起到重要的免疫功能。
Objective To investigate the changes of rats omental type Imilky spots in area, volume and ultrastructure of macrophages in inflammatory status , and to observe the function of type I milky spots. Methods SD rats induced immune response by intraperitoneal injection of Sapylin. Double - labeling of rats omental milky spots with activated carbon and HE staining, measuring the number and the area of type Ⅰ milky spots by Image -pro Plus software, observing the differences in ultrastructure between two groups of type I milky spots byelectron microscope. Results Compared with the control group, the area of the type Ⅰ milky spots in experimental group was significantly larger [ (0. 316 ±0. 066) mm2 vs (0. 102 ±0. 041 )mm2, p 〈0.01 ], the num ber of type Ⅰ milky spots in experimental group was significantly increased [ ( 11. 053 ± 1. 543 ) per cm2 vs ( 1. 913 ±0.851 )per cm2, p 〈0. 01)]. Experimental group had lots of mitochondrias, lysosomes, golgi apparatus and other organelles in macrophages of type Ⅰ milky spots, the rough endoplasmic reticulums were expansion, pseudopods were enlarged and dense. Conclusion Sapylin can activate the macrophages of type Ⅰ milky spots, increase the number and the area of type Ⅰ milky spots. It demonstrate that the type Ⅰ milky spots play an important role in immune protection of abdominal cavity.
