再生障碍性贫血患者外周血T-bet和GATA-3转录因子基因表达失衡的分析 被引量:6

Analysis of Gene Expression Unbalance of T-bet/GATA-3 Associated Transcription Factors in Patients with Aplastic Anemia

摘要 目的探讨再生障碍性贫血(aplastic anemia,AA)患者外周血单个核细胞中T-bet/GATA-3的比率与Th1/Th2细胞失衡的关系。方法采用RT-PCR法检测30例初诊AA患者及20名健康人外周血单个核细胞(PBMC)转录因子T-bet、GATA-3 mRNA的表达水平,ELISA法检测血浆IFN-γ和IL-4水平。结果 AA组PBMC中T-bet mRNA及血浆IFN-γ水平均显著高于正常对照组(P分别<0.01),且T-bet/GATA-3 mRNA的比率较对照组明显升高(P<0.01),Th1类细胞因子IFN-γ的水平与T-bet/GATA-3的比率成正相关(r=0.84),而Th2类细胞因子IL-4的水平降低,与T-bet/GATA-3的比率成负相关(r=0.75)。结论 T-bet和IFN-γ表达的增高在AA的发病中可能起到重要作用;AA患者T-bet/GATA-3的比率升高可作为评价Th1/Th2细胞失衡的另一重要指标,在协助临床对AA诊断中起重要作用。 Objective To explore the relationship between the ratio otT-bet/(JAlA-3 ot transcription tactors ana lnl/ln2 type cytokines in peripheral blood mononuclear cells (PBMCs) in patients with aplastic anemia. Methods The expression of Thl/Th2 type cytokines 1FN-γ/IL-4, transcription factors T-bet/GATA-3 in PBMCs from 30 patients with AA and healthy controls were determined by ELISA and RT-PCR respectively. Results It was showed that T-bet mRNA and Thl type cytokines IFN-γ in patients with AA were higher than those of control group respectively, to opposite, GATA-3 mRNA and Th2 type cytokines IL-4 in patients with AA were lower than those of control group; the ratio of T-bet/GATA-3 mRNA in AA was higher than that of control group. Spearman correlation analysis revealed that the T-bet/GATA-3 mRNA ratio was positively correlated with IFN-y in AA, but negatively with IL-4. Conclusions The increased expression of both T-bet mRNA and Thl type cytokines IFN-y may play an important role in immunopathology of aplastic anemia; the ratio of T-bet/GATA-3 may be considered as another accurate indicator of the imbalanced ratio of Thl/Th2 in patients with AA.
