摘要
目的:观察GABAA-R拮抗剂荷苞牡丹碱(Bicuculline,Bic)及苯二氮(BDZ)受体拮抗剂氟马西尼(Flumazenil,Flu)对解虑安神汤(JLAST)乙酸乙酯提取部位(J-A)和正丁醇提取部位(J-B)抗小鼠焦虑行为的影响,研究JLAST抗焦虑作用与GABA-BDZ-Cl-复合体的关系。方法:小鼠连续给予J-A、J-B 7 d,于末次给药后1 h,脑室内分别注射Bic或Flu,观察J-A、J-B对小鼠高架十字迷宫实验行为学的影响。结果:与空白对照组相比,①地西泮组、J-A组、J-B组均可增加小鼠进入开臂次数、进入开臂次数百分比(OE%)及开臂停留时间百分比(OT%),差异有显著性意义;②地西泮+Bic组、J-A+Bic组、J-B+Bic组对小鼠进入开臂次数、在开臂停留时间及OE%、OT%无明显影响,差异无显著性意义;③地西泮+Flu组、J-A+Flu组对小鼠进入开臂次数、在开臂停留时间及OE%、OT%无明显影响,差异无显著性意义;J-B+Flu组可增加小鼠进入开臂次数、在开臂停留时间百分比(OT%),缩短在闭臂停留时间,差异有显著性意义。结论:JLAST抗焦虑的机制与GABAA-R有关,其中J-A可能通过GABA-BDZ-Cl-复合体途径发挥作用,J-B则可能通过GABAA-R相关的其他途径发挥作用。
Objective: To observe the effects of GABAA-R antagonist(Bicuculline,Bic) and Benzodiazepine(BDZ) and receptor antagonist(Flumazenil,Flu) on the ethyl acetate extracts(J-A) of Jielv Anshen Decoction and the butanol extracts(J-B) of anti-mouse anxiety and to study the relation of JLAST anxiolytic effect and the GABA-BDZ-Cl-complex.Methods: Male mice were administered either with J-A or J-B for 7 days,and then their ventricles were injected with Bic or Flu 1h later after the last administration to observe the impact of JA,JB on the mice behavior in cross elevated maze test.Results: Compared with the non-medicine group and the control group,(1) the diazepam group,J-A Group and J-B Group could increase the times of the mice's opening their arms and the percentage of the mice's opening entry(OE%) and the percentage of the mice's opening time(OT%),with significant difference.(2) The diazepam+Bic group,J-A+Bic group and J-B+Bic group showed no obvious impacts on the times of the mice's opening their arms and the percentage of the mice's opening entry(OE%) and the percentage of the mice's opening time(OT%),with non significant difference.(3) The Diazepam+Flu group and J-A+Flu group showed no obvious impacts on the times of the mice's opening their arms and the percentage of the mice's opening entry(OE%) and the percentage of the mice's opening time(OT%),with non significant difference,but J-B+Flu group could increase the times of the mice's opening their arms and the percentage of the mice's opening entry(OE%) and shorten the time of the mice's stay in closing their arms,with significant difference.Conclusions: JLAST anxiolytic mechanism has something to do with GABAA-R,among which J-A may play via the GABA-BDZ-Cl-complex while J-B may play via the other pathway related to GABAA-R.
