直接竞争酶联免疫吸附分析法测定牛奶中磺胺二甲嘧啶

Direct Competitive Enzyme-Linked Immunosorbent Assay for Suiphadimidine Residues in Milk

应用自制的磺胺二甲嘧啶(SM_2)-蛋白质偶联物,经免疫得高质量兔抗SM_2抗血清。建立了测定SM_2的酶联免疫吸附分析(ELISA)竞争法,重现性好,特异性强,测定线性范围0.05～5.0μg·L^(-1),最低检出浓度为0.025μg·L^(-1)。全脂牛奶与脱脂牛奶的加标实验回收率分别为82.1％～128.8％和90.6％～124.0％。测定了270个牛奶试样,未发现大于欧盟允许的最高浓度25μg·L^(-1)的阳性试样。

High qualitative anti-SM2 serum of rabbit was obtained through immunozation with self-produced sulphadimidine (SM2)-protein conjugates. The direct competitive enzyme-linked immunosorbent assay for SM2 was established, reproducibility being good, specificity potent. The linear range determined was 0.05-5.0 μg·L-1, lowest detection limit was 0.025 μg·L-1. Added standard experimental recoveries of wholl milk and skimmed milk were from 82.1 to 128.8% and from 90.6 to 124.8% , respectively. The positive sample of peak concentration up to 25 μg·L-1 more than the upper limit allowed EU had been not found.
