摘要
目的:探讨溶栓颗粒含药血清对TNF-α损伤血管内皮细胞VEGF表达的影响。方法:用TNF-α损伤大鼠脑皮质微血管内皮细胞(BMECs)造模,用含大、中、小剂量溶栓颗粒和中等剂量阿托伐他汀的大鼠血清干预受损的BMECs,应用HE染色观察细胞形态变化,免疫组化法观察血管内皮细胞生长因子(VEGF)的表达。结果:TNF-α可以造成BMECs损伤,表现细胞数量下降、皱缩;受损细胞的VEGF表达明显增多。含大、中剂量溶栓颗粒和阿托伐他汀的大鼠血清能减少VEGF表达,阿托伐他汀的这一作用与大剂量溶栓颗粒相当。结论:溶栓颗粒可以减少因炎性介质TNF-α增加而受损的血管内皮细胞的VEGF表达,可能是其减缓动脉粥样硬化形成的作用机制之一。
Objective: To discuss the effect of serum containing rongshuan granule on VEGF expression in vascular endothelial cell damaged by TNF-α.Methods: Models were made by applying TNF-α to injury vascular endothelial cells of rats' cerebral cortex micrangium,the serum containing rongshuan granule high,middle,low dosage and atorvastatin was respectively applied to intervene the damaged vascular endothelial cells,the cells' morphological changes were observed by HE staining,the vascular endothelial growth factor(VEGF) expression on the damaged vascular endothelial cells was determined by immunohistochemical method.Results: Vascular endothelial cells were damaged by TNF-α,that showed the number of cells was decreased,cells shrank and the VEGF expression on the vascular endothelial cells was obviously increased.Serum containing rongshuang granule middle,high dosage and atorvastatin could reduced the VEGF expression and improve the morphological changes of vascular endothelial cells,the effect of atorvastatin was correspond to the effect of the high dosage group.Conclusion: Rongshuan granule can decrease the expression of VEGF on vascular endothelial cells damaged by TNF-α,that maybe one of the mechanism that rongshuan granule delay atherosclerosis formation.
