摘要
目的:研究慢性特发性血小板减少性紫癜(CITP)患者外周血CD4+CD25+CD127low调节性T细胞及相关细胞因子的表达水平,探讨它们在ITP发病机制中的作用。方法:流式细胞术(FCM)检测31例ITP患者及25例健康志愿者外周血CD4+CD25+CD127low调节T细胞的数量,ELISA方法检测血清中IL-10、TGFβ-1的含量,并进行相关性分析。结果:ITP患者外周血CD4+CD25+CD127low调节性T细胞数量明显低于正常对照组(P<0.05),血清IL-10、TGFβ-1的含量也较正常对照组低(P<0.05)。CD4+CD25+CD127low调节性T细胞数量与IL-10的表达有相关性(P<0.05),与TGFβ-1的表达水平无相关(P>0.05)。结论:ITP患者CD4+CD25+CD127low调节T细胞数量减少及相关细胞因子的降低可能与ITP的细胞免疫失调有关。
Objective:To investigate the amount of CD4+CD25+CD127low regulatory T cells and related cytokines in peripheral blood of the patients with idiopathic thrombocytopenic purpura(ITP),and discuss the role they play in the pathogenesis of ITP.Methods:The amount of CD4+CD25+CD127low regulatory T cells in the peripheral blood of 31 ITP patients and 25 healthy donors were evaluated by flow cytometry,ELISA was used to test the level of IL-10 and TGF-β1 in blood serum,and then we analyzed the correlation between levels of CD4+CD25+CD127low regulatory T cells and IL-10,TGF-β1.Results:ITP patients had a lower proportion of CD4+CD25+CD127low regulatory T cells than the healthy donors(P〈0.05).The level of IL-10 and TGF-β1 in ITP patients was also lower than that in healthy donors(P〈0.05).There was positive correlation between levels of CD4+CD25+CD127low regulatory T cells and IL-10(P〈0.05).There was no positive correlation between levels of CD4+CD25+CD127low regulatory T cells and TGF-β1(P〉0.05).Conclusion:Decrease of CD4+CD25+CD127low regulatory T cells and related cytokines in ITP patients may be connected with their cellular immunity disturbance.
