摘要
目的观察1-氯甲基杂氮硅三环长期暴露对SD大鼠脑神经丝蛋白表达的影响,探讨其神经毒性作用机制。方法选择4周龄无特定病原体(SPF)级SD大鼠60只,分为对照组和染毒组。染毒组按照雌性4.00 mg.kg-1.d-1、雄性动物3.44 mg.kg-1.d-1的剂量经口饲养12个月后用过氧化酶法检测脑组织中神经丝蛋白200(NF200)的表达变化情况。结果长期摄食1-氯甲基杂氮硅三环可导致雌雄大鼠大脑海马CA2、CA3区神经元核浓染、固缩,神经元数目减少和NF200蛋白表达明显降低。结论海马CA2、CA3区神经元的损伤及NF200蛋白的表达的降低是1-氯甲基杂氮硅三环神经毒性的表现方式之一。
Objective To explore the toxic effect of 1-chloromethylsilatrane long term exposure on high molecular weight neurofilament protein(NF200)expression and mechanism of neurotoxicity of 1-chloromethylsilatrane in SD rats.Methods 60 healthy 4-week old SD rats were randomly divided into 2 groups,a control group and an exposed group.1-Chloromethylsilatrane mixed in formula feeds was administered at the dosages of 4.00 mg·kg-1·d-1 in female and 3.44 mg·kg-1·d-1 in male for 12-month and then NF200 was determined by immunohistochemical technique.Results After a long term exposure of 1-chloromethylsilatrane,the expression level of NF200 was obviously decreased in the CA2 and CA3 of hippocampus,and the damage was found in hippocampal neuron with pyknotic nuclei and abnomally decreased number.Conclusion On this experimental condition,4.00 mg·kg-1·d-1 in female and 3.44 mg·kg-1·d-1 in male,the damage to neurons and the decrease of NF200 in the CA2 and CA3 of hippocampus may be one of the possible toxic mechanism caused by 1-chloromethylsilatrane.
