糖基化磷脂酰肌醇特异性磷脂酶D的表达与白血病细胞黏附和侵袭性的关系

摘要 目的比较髓性白血病细胞K562、HL-60和THP-1黏附、迁移能力以及糖基化磷脂酰肌醇特异性磷脂酶D(GPI-PLD)的表达,探讨GPI-PLD表达与不同白血病细胞黏附性、迁移性的关系。方法采用黏附实验和体外穿膜实验比较3种不同白血病细胞的黏附性和侵袭性;半定量RT-PCR检测这些白血病细胞GPI-PLD和uPAR mRNA的表达水平;免疫印迹法检测细胞GPI-PLD的表达;流式细胞术检测细胞膜uPAR的表达。结果 THP-1细胞中GPI-PLD和uPAR的mRNA及蛋白质表达水平均明显高于K562和HL-60细胞。THP-1细胞对Fn的黏附率显著高于K562、HL-60细胞;THP-1与HL-60细胞体外穿膜侵袭能力显著高于K562细胞。结论 THP-1细胞较K562和HL-60细胞具有较高的黏附和侵袭能力,与其GPI-PLD和锚定蛋白uPAR表达有关。
