摘要
目的观察不同剂量的白藜芦醇对喂饲高脂高胆固醇饲料的小鼠血清和肝脏脂质过氧化水平的影响。方法将50只昆明种小鼠按体质量随机分为基础组(n=10)和高脂组(n=40),分别喂饲基础饲料和高脂高胆固醇饲料。4周后,按体质量和血清总胆固醇水平将高脂组随机分为4组:高脂对照组、白藜芦醇低(5mg·kg-1·d-1)、中(22.5mg·kg-1·d-1)、高(45mg·kg-1·d-1)组。基础组和高脂对照组均给予0.5%羧甲基纤维素钠溶液灌胃;白藜芦醇低、中、高组小鼠分别经灌胃给予相应剂量的白藜芦醇羧甲基纤维素钠溶液。干预6周后测定小鼠血清和肝脏中谷胱甘肽过氧化物酶、超氧化物歧化酶、丙二醛和总抗氧化能力的水平。结果基础组、白藜芦醇低、中、高组小鼠肝脏丙二醛水平均低于高脂对照组小鼠(P<0.05)。结论白藜芦醇能够抑制喂饲高脂高胆固醇饲料小鼠肝脏中丙二醛水平的升高。
Objective To study the effects of resveratrol on lipid peroxidation in mice with high fat and high cholesterol diet.Methods Fifty mice were randomly assigned to the following two groups: the mice of control group (n =10) were given a normal diet,and the mice in high-fat diet group (n =40) given high fat diet for 4 weeks.And then the mice in high-fat diet group were randomly assigned to the following four groups according to body weight and serum total cholesterol level: high-fat diet control group;low-dose resveratrol group(5 mg·kg-1·d-1);intermediate-dose resveratrol group(22.5 mg·kg-1·d-1);high-dose resveratrol group(45 mg·kg-1·d-1).The resveratrol was dissolved in 0.5% sodium carboxymethyl cellulose and given by intragastric administration.The levels of glutathione peroxidase (GSH-Px),superoxide dismutase (SOD),malondialdehyde (MDA),total antioxidant capacity (T-AOC) were measured after the treatment lasted for 6 weeks.Results The MDA levels in high-fat diet control group are higher than those in other groups (P 〈 0.05).Conclusion Resveratrol (545 mg/kg·d-1) could decrease MDA levels in liver of mice with high fat and high cholesterol diet.
