缺氧诱导因子1α在宫内缺氧缺血性脑损伤的胎鼠脑组织中的表达 被引量:2

The Expression of Hypoxia Inducible Factor 1 Alpha in Fetal Rat Brain Tissue Injured by Intrauterine Hypoxia-ischemia

摘要 目的探讨宫内缺氧缺血性脑损伤后不同复氧时间胎鼠脑组织中缺氧诱导因子1α(HIF-1α)蛋白的表达规律。方法孕19d Wistar大鼠36只,随机分为对照组和8个实验组,每组4只。实验组建立宫内缺氧缺血性脑损伤动物模型后,分别于子宫恢复血供后0、0.5、2、4、8、12、24及48h取胎鼠脑组织;对照组只暴露双侧子宫15min后,取胎鼠脑组织,应用免疫组织化学方法检测HIF-1α蛋白的表达。结果实验组缺氧诱导因子1α蛋白表达在术后8h明显升高,12h达到高峰,24h后明显下降。结论 HIF-1α在宫内缺氧缺血性脑损伤的胎鼠脑组织中的表达随着复氧时间的延长呈现先升高后降低的趋势。 Objective To investigate the expression pattern of hypoxia inducible factor 1 alpha (HIF-1α) in fetal rats′ brain tissue injured by intrauterine hypoxia-ischemia.Methods Fetal rats were obtained from 36 pregnant Wistar rats at the gestational 19 days.The 36 pregnant rats were randomly divided into 1 control group and 8 experimental groups.The pregnant rats of experimental groups were modeled into intrauterine hypoxia-ischemic brain injury,and then brain tissue of fetal rats were obtained at 0 h,0.5 h,2 h,4 h,8 h,12 h,24 h and 48 h after hypoxia,respectively.The pregnant rats in control group were not modeled and brain tissues of fetal rats were obtained directly.HIF-1α was examined by S-P immunohistochemistry.Results The HIF-1α expression was gradually increased until 8 h,reached maximal level after 12 h and recovered to basic level by 24 h.Conclusion The expression of HIF-1α in fetal rats′ brain tissue undergoes gradual increase and decrease after intrauterine hypoxia-ischemic injury.
