肺鳞癌、腺癌CD44v、P16基因蛋白表达与其生物学行为之间关系研究 被引量:3

摘要 目的 探讨肺鳞癌、腺癌CD44v、P16基因蛋白表达与其生物学行为之间关系。方法 采用链霉菌抗生物素蛋白—过氧化物酶(SP)免疫组织化学法检测78 例非小细胞肺癌患者肺癌组织CD44v6和P16的表达。结果 CD44v6在正常支气管粘膜表达阴性,鳞癌和腺癌患者的阳性表达分别为50% (18/36)、57.1% (24/42,P> 0.05)。有淋巴结转移组的阳性表达高于无转移组分别68.18%(30/44)35.29% (12/34,P< 0.01)。阳性表达的患者3年生存率为24% ,明显低于阴性者(67% ,P<0.01)。P16 基因蛋白在16例正常支气管粘膜上皮细胞的表达为94.0% ,鳞癌和腺癌患者的阳性表达分别为55% (20/36),66.66% (28/42,P> 0.05)。78 例肺癌P16 基因蛋白的阳性表达率为61.53% ,正常支气管粘膜与肺癌之间的P16基因蛋白阳性表达率差异显著(P< 0.01)。有淋巴结转移组的阳性表达为32% (14/44),低于无转移组97% (33/34,P< 0.05)。阳性表达患者的3 年生存率为84% ,明显高于阴性患者27% ,(P< 0.01)。P16蛋白表达随着肺? ? Objective To study the expressions of CD44v6、P16 proteins and their correlation with biologic behaviors on human lung squamous cancer and adenocarcinoma.Methods A rapid immunohistochemical method (streptoavidin peroxidase conjugated method,SP) was used to detect CD44v6、P16 proteins expressions in lung cancer.Results Expression of CD44v6 was negative in normal bronchial mucosa,50%(18/36) in squamous cell carcinoma(SCCs),57.1%(24/42) in adenocarcinomas(ACs)(P>0.05).The incidence of CD44v6 in patients with lymphaden metastasis was 68.18%,which was significantly higher than that without lymphaden metastasis(35.29%.P<0.01).The 3 year survival rate of the patients with CD44v6 positive tumors was 24%,which was significantly lower than that of CD44v6 negative patients(67%,P<0.01).The expression of P16 was 94% in normal bronchial epithelial cell,55%(20/36) in SCCs,66.66%(28/42) in Acs(P>0.05).The incidence of P16 patients with lymphaden metastasis was 32%(14/44),which was significantly lower than that without lymphaden metastasis(97%,P<0.05).The 3 year survival rate of the patients with P16 positive tumors was 84%,which was significantly higher than that of P16 negative patients(27%,P<0.01).The inactivation of P16 protein was implicated in tumor differentiation.Conclusion CD44v6 and P16 were two independent factors involved in positive negative regulating in carcinogenesis, wich were important markers of metastasis and prognosis in NSCLC. 〔
