锁阳中儿茶素、根皮苷的提取及含量测定 被引量:10

Extraction and Determination of(+)-Catechin and Phloridzin from Cynomorium Songaricum

摘要 建立了高效液相色谱―二极管阵列检测器联用测定中药锁阳中黄酮类化合物儿茶素、根皮苷的方法.用C18反相色谱柱梯度洗脱法成功分离了二者;以儿茶素和根皮苷含量总和为指标,以物料比、提取时间和提取次数为正交试验考察因素,优化了锁阳中黄酮的提取方法,得到的最佳提取条件为:28 kHz、50℃条件下,以药材量7.5倍的甲醇超声提取3次,每次30 min.该方法提取效果好,精密度、准确度和稳定性较好,用于11个不同产地锁阳样品的提取和含量测定,可作为锁阳及其制品中儿茶素和根皮苷等黄酮类化合物提取、分析及质量控制的方法. A method for the simultaneous determination of (+)-Catechin and Phloridzin in extracts of Cynomoriurn songsrieum was developed using high performance liquid chromatographic (HPLC) coupled with diode array detector (DAD). A good separation was achieved by using a C^s HPLC column with gradient elution. At the same time, the method for extracting flavonoids from C. songsricum was optimized using the total content of (+)-Catechin and Phloridzin as extraction index and with the ratio of material to liquid, extraction duration and extraction times as influential factors in the orthogonal experiment. The optimum extracting conditions were 7.5 times quantity of methanol to herbal material at 28 kHz and 50 ℃, extraction for three times for 30 min each time. The proposed method was used to extract and determine the two flavonoids in C. songaricum collected from different producing area. The result showed that the method was good in aspects of extraction, precision, accuracy and stability. It can provide a reference to the extraction, analysis and quality control of flavonoids in C. songaricum and its products.
