摘要
目的:探讨骨髓间充质干细胞(BMSCs)对慢性阻塞性肺疾病(COPD)大鼠的治疗作用。方法:健康雄性SD大鼠,随机分成3组,正常对照组、模型组和治疗组。通过烟熏法建立COPD模型后,大鼠尾静脉注射BMSCs进行生物治疗。应用细胞膜荧光染料PKH26标记的BMSCs回输大鼠体内,制作肺组织冰冻切片,在荧光显微镜下观察BMSCs是否定位于肺组织;采用硫代巴比妥酸和黄嘌呤氧化法分别检测血清和肺组织中超氧化物歧化酶(SOD)和活性丙二醛(MDA)含量;收集支气管肺泡灌洗液(BALF),计数细胞总数,并进行分类;制作肺组织病理切片,观察肺组织微细结构变化并定量测定肺平均内衬间隔(MLI)、平均肺泡数(MAN)。结果:BMSCs能够归巢到COPD大鼠的肺组织并存活;BM-SCs治疗组与模型组相比,血清中SOD的活性明显增高、MDA含量显著降低(P<0.05);BALF中细胞总数明显减少,中性粒细胞总数及比例均减小(P<0.05);病理有明显的改善,MLI减小,MAN增大(P<0.05)。结论:BMSCs对COPD大鼠具有治疗作用。
Objective:Aim to study the therapeutical effect of bone marrow mesenchymal stem cells(BMSCs) on rats chronic obstructive pulmonary disease(COPD) model.Methods:Fifteen male SD rats were randomly divided into a normal control group,a model group and cell transplantation group.Rat models of COPD were established by exposure to cigarette smoking daily,80 days later,in the cell transplantation group,1 mL cell suspension was infused into the rat caudal vein,containing 3 × 106 BMSCs,totally four times.Ani-mals were received BMSCs marked with PKH26 to detect weather MSCs can reach lung tissue,detected serum and lung malondialdehyde(MDA) content and superoxide dismutase(SOD) activity,collected bronchoalveolar lavage fluid(BALF),counted total cell number,and classified,lung tissure section stained by hematoxylin and eosin(HE)was observed to study the morphological alterations.Mean linear intercept(MLI) and mean alveo-lar numbers(MAN) were also measured.Results:BMSCs can home to lung of COPD on rat models,oxygen free radical examination results showed that compared with the model group,SOD activity was significantly higher(P 0.05),MDA levels were significantly lower(P 0.05) in the cell transplantation group.compare with the model group,the cell transplantation group the total cell and neutrophil counts in bronchoalveolar lav-age fluid(BALF) were decreased(P 0.05),MLI was higher in model group than in the cell transplantation group,while MAN were on the contrary(P 0.05).Conclusion:BMSCs transplantation can effectively treat rat models of COPD.
