摘要
采用HPLC法测定硝酸咪康唑脂质体中硝酸咪康唑含量和包封率。采用C18柱,流动相为0.5%醋酸铵溶液-乙腈-甲醇(15∶42.5∶42.5),检测波长230 nm,流速1 mL/min,采用Sephadex LH-20柱分离脂质体与游离药物。结果表明,硝酸咪康唑在0.25~15μg时,线性关系良好(r=0.999 1),回收率为99.67%(n=9),所测脂质体的平均包封率为75.54%。该法准确,重现性好,可用于测定硝酸咪康唑脂质体的药物含量及包封率。
A HPLC method for content and entrapment efficiency determination of miconazole nitrate liposome was established.Chromatographic column was C18 column,mobile phase was 0.5% ammonium acetate solution-acetonitrile-methanol(15 ∶42.5 ∶42.5),the wave length was 230 nm,and the flow rate was 1 mL/min.The liposome was separated from free drug with sephadex LH-20.Results showed that miconazole nitrate had a good linear relation over the range of 0.25~15 μg(r=0.999 1),the recovery was 99.67%(n=9).The average entrapment efficiency was 75.54%.This method is accurate,reproducible and applicable for determination of content and entrapment efficiency of miconazole nitrate liposome.
