摘要
目的研究金丝桃苷(hyperfine,HP)对四氯化碳(CCl4)诱导肝细胞损伤的保护作用及其机制。方法用CCl4体外诱导人正常肝细胞L-02损伤,检测培养上清液中天门冬酸氨基转移酶(AST)和丙氨酸氨基转移酶(ALT)水平,测定肝细胞中丙二醛(MDA)和谷胱甘肽(GSH)的含量,四甲基偶氮唑盐微量酶反应比色(MTT)法检测肝细胞存活率。结果金丝桃苷可明显降低由CCl4升高的肝细胞培养上清液中ALT和AST水平及肝细胞MDA含量,还可提高由CCl4降低的肝细胞存活率和GSH含量。结论金丝桃苷对人肝细胞氧化性损伤有直接保护作用,这可能与金丝桃苷抑制抗氧化酶活性和抗自由基活性有关。
Objective To study the protective effect and the mechanism of hyperfine(HP) on liver cell injure induced by CCl4.Methods CCl4 was used to induce the human liver cells L-02 injury in vitro.The contents of malondialdehyde(MDA) and glutathione(GSH)in liver cells and the levels of AST and ALT in cultural supernatant were determined by general methods.Cell viability was assayed by MTT method.Results The elevation of MDA content in liver cells and AST and ALT levels in supernatant of cultural cells,and the decrease in cell viability and GSH content induced by CCl4 were restored remarkably by the treatment with hyperine.Conclusion Hyperine possesses the protective effect on the liver injured induced by CCl4 in vitro,which might be associated with the anti-oxidative and anti free radicals activities of hyperine.
