高效液相色谱法测定归芍调经胶囊中芍药苷的含量

Determination of paeoniflorin in Guishao tiaojing capsules by high performance liquid chromatography

目的应用高效液相色谱法测定归芍调经胶囊中芍药苷的含量。方法色谱柱:Diamonsil C18柱(150 mm×4.6 mm,5μm);柱温:30℃;流动相:乙腈-0.1%磷酸溶液(16∶84);检测波长230 nm;流速:1.0 ml.min-1。样品处理过程中,我们对提取溶媒,提取方式,以及提取时间进行简单的对比筛选。最后选择甲醇作溶媒;提取方式为超声提取;提取时间40 min。结果芍药苷在60.45～362.7 mg.L-1范围内,其浓度与峰面积线性关系良好,回归系数r=0.999 9(n=6),本方法的平均回收率为100.19%,RSD为1.24%。结论该方法稳定可靠,重现性好,可有效控制归芍调经胶囊的质量。

Aim To establish a method of determing paeoniflorin in the Guishao tiaojing capsules by high performance liquid chromatography.Method The HPLC method was used with Diamonsil C18 column（150 mm×4.6 mm,5 μm）.The column temperature was 30℃.The mobile phase was acctonitrile-0.1%phosphoric acid（16∶ 84）.UV detection wavelength was set at 230 nm and the flow rate was 1.0 ml·min-1.We compared and screened ertraction solvent,extraction method,and the extraction time.Methanol was selected as solvent,with ultrasonic extraction as method and the extraction time was 40 min.Results Within the range of 60.45 ~ 362.7 mg·L-1,paeoniflorin＇s concentration to peak linear was good.The regression coefficient was r=0.999 9（n= 6）.The average recovery rate was 100.19 % and RSD was 1.24 %.Conclusion This method is stable,reliable and reproducible.It can be used for the quality control of Guishao tiaojing capsule.
